Aspects of the immune response against proteic antigens submitted to the effects of 60Co gamma radiationConsidering the effects of gamma radiation on proteins and the capacityof the immune system to recognize modified macromolecules, we decidedto evaluate some immunological Aspects of B10.PL mice exposed to nativeor irradiated ovalbumin and bothropstoxin-1 (BthTx-1). In order toevaluate possible structural modifications of the molecules after beingirradiated (60Co gamma rays), bothropstoxin-1 was analysed byelectrophoresis, while ovalbumin was submitted to analytical sizeexclusion chromatography. The toxin was also analised by ESI-massspectrometry. Our results indicate that radiation promotedmodifications on both the molecules. Aiming to compare the toxicity ofthe native and irradiated forms of the toxin, an in vitro citotoxicityassay, using CHO cells, was performed. According to our results, themodified toxin was 5 folds less toxic than its native counterpart. Seraof animals immunized with the native and irradiated proteins wereanalyzed in order to evaluate levels of IgG, as well as to quantifyspecific isotypes. While the native proteins induced a predominant Th2response, the irradiated molecules apparently promoted a switch towardsa Th1 pattern. We also performed a cell proliferation assay withsplenocites from mice immunized with either the native or theirradiated proteins, cultured in the presence of the antigens. Ourresults indicate that both the forms of the proteins induced a similarproliferative response. These data indicate a potential use ofdetoxified proteins as antigens for immunization.
