When MI6 operative John Strangways and his secretary mysteriously disappear, James Bond is sent to Jamaica to investigate whether their disappearance is related to the recent disruptions of American missile launchings.

On arrival in Jamaica, it soon becomes apparent that Strangways and his secretary have been murdered. Bond learns that Strangways had been investigating the activities of Dr. Julius No, a reclusive Chinese-German living on Crab Key, an island rumoured to be roamed by a vicious dragon.

With Strangways' classified information on Dr.No's operations missing, Bond learns that the seemingly respectable Professor Dent was Strangways' last contact before his disappearance. But Dent is an agent of Dr. No with orders to kill Bond after he discovers radioactive rock samples originating from Crab Key. Dent is unsuccessful in his attempt to kill Bond and, after a brief interrogation in which Bond confirms Strangways is dead, Bond executes Dent.

With the help of CIA operative Felix Leiter and his Jamaican contact Quarrel, Bond makes plans to investigate Crab Key island. As Bond and Quarrel sail to Crab Key, Leiter remains behind to alert the authorities should Bond fail to complete his mission within the next twelve hours.

They arrive undetected, but the following morning Bond meets an unexpected visitor to Crab Key, the beautiful Honey Ryder. With Ryder in tow, Bond and Quarrel search the island while trying to avoid Dr. No's private army who scour the island for the unwelcome guests. Bond and Ryder are eventually captured by Dr. No's army, but Quarrel is burned to death by the feared 'dragon' of Crab Key.

While held captive in Dr. No's private headquarters, Bond learns that Dr. No, who ostensibly operates a business harvesting and exporting guano, is in fact an agent of SPECTRE working with the Russians. They have supplied him with several million dollars worth of equipment to sabotage nearby American missile tests.

Bond also learns first hand that Dr. No's speciality is torture. Dr. No inflicts punishment on Bond to discover and record his powers of endurance. But Bond survives, and soon finds himself avoiding death while crawling through a red-hot ventilator shaft en-route to the main control room.

Once inside, Bond and Dr. No confront each other atop the vat of heavy water, into which Dr. No soon succomes. Bond then overloads the control room's nuclear reactor and escapes the island with Ryder just in time!
